10x YNB stock (100 ml): 6.7 g YNB w/o amino acids + carbon/energy source

if you don't use amino acids as selection marker, use YNB+amino acids for faster growth

10x YNB + glucose (100 ml): 6.7 g YNB, 20 g glucose

filtersterilize or autoclave for 10 min.


YPD

(yeast and E. coli)

1% yeast extract
2% bactopeptone
2% glucose

(add 2% agar for plates)

autoclave for 10 min.


2YT

(E. coli and yeast)

1.6% bactotryptone
1% yeast extract
0.5% NaCl

minimal media for E. coli

MOPS buffered minimal medium for E. coli and S. typhimurium (adapted from Neidhart et al.(1974) J Bacteriol 119, 736-47)

for 1 liter:

100 ml 10x MS
1 ml 1.32 M K2HPO4
40 ml 1M MOPS pH 7.4
10 ml 50% glucose
2.5 ml 4 mg/ml thiamine
amino acids (at 4 mg/ml = 100x), antibiotics etc. as required (for MC1000-derived strains: valine, leucine, isoleucine, thiamine)

10x MS and K2HPO4 can be autoclaved, the rest should be filtersterilized

for 1 liter 10x MS:
40 ml 1 M Tricine (freshly made), pH 7.4 with 50% KOH
50 ml 1.9 M NH4Cl
10 ml 0.276 M K2SO4
10 ml 10 mM FeSO4 (freshly made)
10 ul (!) 0.5 M CaCl2
5.3 ml 1 M MgCl2
100 ml 5 M NaCl
100 ul (!) 10.000x micronutrients

tricine and FeSO4 should be filtersterilized, the rest can be autoclaved

for 10.000x micronutrients:
