antibiotics stock solutions

ampicillin

1000 x stock = 50 mg/ml in water

use as 500 x stock for high-copy plasmid-containing cells on rich-media plates to reduce satelite growth

stock can be prepared in 70% ethanol to keep it liquid in the freezer. dissolve ampiccilin in 0.3 vol water (use a few drops of concentrated HCl to dissolve the powder), then add ethanol

interferes with cell wall synthesis


chloramphenicol

1000 x stock = 35 mg/ml in ethanol
you can put 50 mg/ml (or more) in ethanol, and up to 25 mg/ml in water

use as 250 x stock for high copy plasmids, as 2000 x stock for single-copy chromosomal inserts

prevents peptide bond formation on ribosomes


kanamycin

500 x stock = 10 mg/ml in water

filtersterilize

causes misreading of mRNA by ribosomes


streptomycin

1000 x stock = 30 mg/ml in water

filtersterilize

causes misreading of mRNA by ribosomes


tetracyclin

1000 x stock = 10 mg/ml in ethanol

light-sensitive

Mg kills tetracyclin, so use a Mg-free medium like 2YT or LB. do NOT use SOB or SOC

prevents protein synthesis by ribosomes
