- grow 2 ml o/n culture in 10ml tube
- spin down 1.5 ml, top speed for 1 min. (remaining 0.5 ml can be stored at 4 C for months)
- remove sup
- vortex pellet into 100 ul ice-cold GET
- 5' room temperature
- add 200 ul frehly made 0.1 M NaOH/1 % SDS, mix GENTLY
- 5' on ice
- add 150 ul ice-cold 3M KOAc pH 4.8
- 5' on ice
- spin 3'
- transfer 400 ul sup to fresh tube. avoid the white stuff!
- add 1 ml EtOH
- 15' -20 C
- spin 5' at top speed, remove sup
- rinse pellet with 70% EtOH (cold), spin 2' at top speed, remove sup
- dry pellet 5' at 37 C or 15' at rt
- dissolve pellet in your favorite buffer (optional: add RNase A)

yield about 1-4 ug, depending on size and copy number of your plasmid

50 mM glucose

3 M KOAc pH 4.8

6 ml 5 M KOAc
1.15 ml acetic acid
2.85 ml water

this is 3M K+ and 5 M -OAc
